Our PhD student Pauline Pfuderer gave a great talk at the London Calling conference run by Oxford Nanopore Technologies on her recent work with Julian Sale's lab at the MRC LMB. Together with Jędrzej Jaworski, Pauline co-led research showing that the replication of extrachromosomal DNA is disorganised: it shows asynchronous replication throughout S-phase, a re-distribution of replication origins, and replication forks that move slowly and are prone to stalling. We're particularly excited about this due to its potential to guide the use of agents that target the DNA damage response in order to selectively disrupt the replication of oncogene-containing extrachromosomal DNA. You can read more about this work in our recent preprint [bioRxiv].
